Annotation of rs3745274
Genotypes GT + TT is associated with concentrations of efavirenz in people with HIV Infections as compared to genotype GG.
Multivariate analysis of factors influencing efavirenz concentrations over 32 weeks of antiretroviral treatment described CYP2B6*6 (516G>T, rs3745274), CYP2B6*11 (136A>G, rs35303484) and ABCB1 rs3842A>G genotypes were consistent significant predictors of plasma efavirenz concentration (see supplemental Table 1). The study also showed that long-term efavirenz auto-induction was more pronounced in patients with the CYP2B*1/*1 genotype compared to those carrying rs3745274 allele T and/or rs35303484 allele G. The study states that the CYP2B6*6, *11 and ABCB1 rs3842A>G genotypes were associated with higher efavirenz plasma concentrations.

Population description
Study Cohort: Anti-retroviral-naive patients with (n=106) or without (n=157) concomitant TB co-infection were treated with efavirenz, zidovudine and lamivudine. Those also being treated for TB were on a rifampicin-based regimen (2 months of ethambutol, isoniazid and rifampicin followed by 4 months of isoniazid and rifampicin).
Note: Alleles in PharmGKB are mapped to the positive chromosomal strand. Therefore, variants in genes on the "minus" strand (eg. VKORC1) are complemented in PharmGKB annotations.
