Pharmacogenetics of Membrane Transporters

Abstract (July, 2005 Update)

Goals

The Pharmacogenetics of Membrane Transporters project (PMT) studies genetic variants of membrane transporters that play a role in drug response pathways. Our proposed studies are focused on two major superfamilies of transporters: SoLute Carrier (SLC) and ATP Binding Cassette (ABC).

Progress

To date, PMT has discovered variants in over 40 membrane transporters and functionally characterized over 80 amino acid variants in 11 membrane transporters. These hypothesis generating studies have provided a foundation for studies of genetic variation in drug response.

Experimental Plans

In our renewal studies, our specific aims are to:

  1. Identify sequence variants in 125 membrane transporter genes in ethnically diverse DNA samples;
  2. Determine cellular phenotypes for transporter variants;
  3. Determine the biological relevance of variants in membrane transporters to clinical drug response;
  4. Deposit the data in PharmGKB and develop shared resources.

Several genotype to phenotype clinical studies are proposed that link genetic variants identified in our cellular assays to clinical drug response. In addition, our phenotype to genotype studies involve GRAD, the Genetics of Response to Anti-Depressants, a rich resource that has accrued over 1000 individuals and contains information about response and adverse drug response to anti-depressants.
